في العقد الأخير، برزت مقاومة المضادات الحيوية كعائق رئيسي أمام العلاج الناجح لالتهابات المسالك البولية، مما استدعى البحث عن استراتيجيات علاجية بديلة. تُعدّ النباتات الطبية والعطرية، مثل الزعتر الشائع، مصدرًا حيويًا للمركبات النشطة بيولوجيًا ذات الإمكانات الدوائية الموثقة، لا سيما في المناطق النامية. هدفت هذه الدراسة إلى فحص التركيب الكيميائي النباتي والفعالية المضادة للبكتيريا للمستخلصات المائية والإيثانولية لنبات T. vulgaris ضد مجموعة سريرية من مسببات الأمراض البولية المعزولة من مريضات في مدينة رداع، اليمن.
أكد الفحص الكيميائي النباتي وجود مجموعة متنوعة من المستقلبات الثانوية، بما في ذلك الفينولات، والفلافونويدات، والتربينويدات، والتانينات، والقلويدات. تم تقييم فعالية المستخلص الإيثانولي كمضاد للبكتيريا باستخدام طريقة الانتشار في الآبار على وسط الآغار، مما أظهر نشاطًا تثبيطيًا واضحًا يعتمد على الجرعة في جميع السلالات المختبرة. فعند تركيز 100 ملغم/مل، أظهر المستخلص الإيثانولي فعالية فائقة، حيث لوحظ أقوى تثبيط ضد S. aureus (29.50 ± 0.50مم) ، تليها E. coli (25.57 ± 0.38 مم) ، بينما أظهرت K. pneumoniae أقل حساسية نسبية.

Antibacterial Activity of Thymus vulgaris Extract against Bacteria Causing Urinary Tract Infections

Abstract
In the last decade, the escalating rate of antimicrobial resistance (AMR) has emerged as a critical barrier to the successful treatment of urinary tract infections (UTIs), necessitating the exploration of alternative therapeutic strategies. Medicinal and aromatic plants, such as Thymus vulgaris, represent a vital source of bioactive compounds with documented pharmacological potential, particularly in developing regions. This study investigated the phytochemical profile and antibacterial efficacy of aqueous and ethanolic extracts of T. vulgaris against a clinical panel of uropathogens isolated from female patients in Rada'a Town, Yemen.
The phytochemical screening confirmed the presence of diverse secondary metabolites, including phenols, flavonoids, terpenoids, tannins, and alkaloids. The antibacterial efficacy was assessed using the agar well diffusion method, demonstrating a clear dose-dependent inhibitory activity across all tested strains. At a concentration of 100 mg/mL, the ethanolic extract exhibited superior potency, with the strongest inhibition observed against Staphylococcus aureus (29.50 ± 0.50), followed by Escherichia coli (25.57 ± 0.38 mm), while Klebsiella pneumoniae showed the lowest relative susceptibility.
Furthermore, the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) values were determined, with the ethanolic extract displaying a high potency (MIC of 12.5 mg/mL for S. aureus), supporting its bactericidal potential. These findings highlight the significant application of T. vulgaris as antibacterial agent.
